Barbie and several young children have traveled to a wide meadow, to help in trees planting. Emma, one of the kindergarten students, comes a cross a small sapling and is determined to plant it, but becomes discouraged when her friends make fun of her for it. Barbie decides to encourage her, saying that even the tallest trees start out as tiny saplings. Barbie has the children gather round while she tells them the story of Thumbelina.
Thumbelina is a Twillerbee, a being who has the power to speed up plant growth, or even great plants from thin air. She is thrilled to learn that a new batch of Twillerbabies is about to arrive, so she creates wings for her two friends, Janessa and Chrysella and herself, intending that the three make use of the wings when witnessing the Twillerbuds bloom into Twillerbabies. Suddenly a convoy of tractors appears on their meadow, and find themselves trapped in a bunch of flowers that are carried to a distant apartment, occupied by Makena, a wealthy and spoiled child and her parents.
After the flower bed is put into Makena’s bedroom, Thumbelina, Chrysella, and Janessa start looking for a way to get back home, but they are chased around by Poofles, Makena’s dog, and are forced to hide again. Makena returns to her bedroom unaware of the Twillerbees and chats on her cellphone with her friend Violet. Apparently her parents are planning on constructing a factory on the same meadow the Twillerbees call home.
Thumbelina come out from hiding to scold the shocked Makena. The girl’s surprise wears off quickly, and she shows off the Twillerbee to Violet, with whom she is very competitive. Afterwards, the trio attempt to get away from Makena and her dog.
Thumbelina decides to send Chrysella and Janessa back to their field to disrupt the construction work while Thumbelina discusses the whole thing with Makena, to persuade her parent to stop the work and extracts a promise from the girl to not tell anyone else about her and the other Twillerbees, offering in return to fashion special things for Makena.
Makena doesn’t take her agreement with Thumbelina seriously and breaks her promise after inviting over Violet and Ashlynn to show them that she is the only one in the world with such a special creature. Furious at the girl, Thumbelina storms off. Violet and Ashlynn prove they are not her true friends by belittling her, and Makena realizes that she has treated Thumbelina badly too.
Deciding to make it up to Thumbelina and save the Twillerbees, Makena heads out to the field to beg Thumbelina’s forgiveness, and convince them that she genuinely wants to save their meadow. Accepting her earnest apology, Thumbelina introduces the girl to the other Twillerbees and shows her the Twillerbuds.
During the evening Makena finds herself happily hard at work in a greenhouse with the Twillerbees her dog and a bird called Lola growing the plants. The following day, she calls her parents over to cancel the factory construction. Thumbelina reveals herself and explains to the girl’s parents, Evan and Vanessa, that the Twillerbuds that are about to blossom create Twillerbabies who grow up into Twillerbees like her. Persuaded to save the Twillerbees as well, Makena’s parent agree to help stop Myron the contractor and keep him from destroying the Twillerbee meadow.
Myron, however, hates flowers and isn’t dissuaded, even when his workers are driven off their work by the birds and Twillerbees, thinking the field is haunted. Finally, Makena, with Thumbelina’s help, puts a stop to Myron’s destructive actions, followed by her parent’s arrival and their shutting down of the factory project. Myron is ultimately chased away by the birds, leaving the girl and her parents at peace to watch the beautiful Twillerbuds bloom. To make sure that the Twillerbees are never again threatened, the meadow is turned into a protected nature park.
And this is in fact the park that Barbie and the children are planting trees in. At the stories end Barbie notes that even the littlest person, and smallest sapling, can make the greatest difference.
Unbeknownst to the children, Thumbelina, Janessa, and Chrysella have been watching Barbie tell the story from a nearby tree. Barbie waves to them. Utilizing their Twillerbee powers, they cause Emma’s tree to grow, showing that everything that Barbie told her students was true.
